Het zal gaan tijd worden dat er eens wat definitief nieuws en of data gemeld gaat worden bij ARWR.
Ondertussen heb ik behoorlijk volgend aankooporder klaargezet voor $ 26,35.
Ik gok er enkel wat op dat we de $22 niet hoeven te zien de komende weken/maanden maar dat is pure gok.

Op Discord leuke samenvatting van % (tot 50%) dat het al mag stijgen om ...gewoon op slotkoers van enkele maanden geleden uit te komen.
Laten we vooral maar hopen dat het momenteel maar enkel short activiteiten zijn.
Dan is het een louter technische wijze om deze rijkgevulde pipeline aan mildere prijs te kunnen bijkopen.

Alles leuk en aardig, maar ik zie alleen een dalende koers. Komt het ooit nog weer goed???
NIEMAND kan een koers voorspellen. Dat heb ik al vaak genoeg meegemaakt bij andere aandelen.

Tegenwoordig is het toch wel vrij simpel om te voorspellen dat we morgen alweer wat dieper gaan dalen.
Evengoed om volgend jaar opnieuw de $40 terug te zien.
Maar toegegeven ,er zijn hierover geen zekerheden.
Kwestie van rustig te vertrouwen in de technologie die ARWR gebruikt.
A propos : aankooporder zonet veranderd naar $26,45 ivm bestaande gap (DSCRD channel)

Wie vandaag het schip nog verlaat kan ik tegen huidige koersen niet zo goed begrijpen.
Er zijn wel degelijk overreacties op Discord.

Mats, jij koos er wel al maanden geleden voor om verder te spreiden naar andere sectoren.
Een goeie keuze, zo blijkt.
Vandaag echter blijf ik zelf pal achter mijn strategie staan.
Als er bloed vloeit door de straten koop ik zelf nog stuk harder bij dan de afgelopen jaren.
Ik zie dit als een buitenkans om nog meer in te kopen dan ik ooit had kunnen voorzien maar het is zeker niet zonder risico's.
Met dank aan de shorters dan maar ...

Huidige koers is niet gebaseerd op data uitslagen, wel op (tijdelijke) onzekerheden.

helaas is die tijdelijke onzekerheid er al 3 a 4 jaar. Dat is wat me toch heel sceptisch maakt op dit moment.

Uw bemerking geeft de huidige koers aan.
De markt heeft altijd gelijk ?
Er heerst op dit moment vrees,ongeduld, onduidelijkheid en behoorlijk wat verhoogd shortersgeweld.
6% shorts ipv 3 à 4% jaartje geleden.
De winnaar zal diegene zijn die deze periode hier juist heeft inschat.
Kan dus perfect de uitstapper ,zittenblijver, bijkoper of shorter worden.

Inclusion Criteria:
Normal pulmonary function tests at Screening (NHVs only)
Confirmed diagnosis of asthma based on source verifiable medical record (asthma patients only)
Confirmed diagnosis of Cystic Fibrosis based on source verifiable medical record (CF patients only)
No abnormal finding of clinical relevance at Screening (other than asthma for asthma patients and CF for CF patients)
Stable dose of asthma controller medications for at least 4 weeks prior to Screening (asthma patients only)
If on allergen-specific immunotherapy, participants must be on a stable maintenance dose
Stable regimen of CF therapy for at least 6 weeks prior to Screening (CF patients only)

Brief Summary: The purpose of this study is to evaluate the safety, tolerability, and pharmacokinetics of ARO-RAGE in normal healthy volunteers (NHVs) and in participants with inflammatory lung disease.. disease (asthma, cystic fibrosis [CF]). In Part 1 of the study, NHVs will receive a single dose of ARO-RAGE or placebo. In Part 2 of the study, adult participants with inflammatory lung disease will receive 2 doses of ARO-RAGE or placebo. Additional NHVs may be randomized to receive 1 or 2 doses of ARO-RAGE or placebo at Sponsor discretion. Dose levels in Part 2 will be determined based on cumulative safety and pharmacodynamic data from Part 1.

Inclusiecriteria:
Normale longfunctietests bij screening (alleen NHV's)
Bevestigde diagnose van astma op basis van een door de bron gecontroleerd medisch dossier (alleen astmapatiënten)
Bevestigde diagnose Cystic Fibrosis op basis van een door de bron te verifiëren medisch dossier (alleen CF-patiënten)
Geen abnormale bevinding van klinisch belang bij screening (behalve astma voor astmapatiënten en CF voor CF-patiënten)
Stabiele dosis astmabeheersende medicatie gedurende ten minste 4 weken voorafgaand aan de screening (alleen astmapatiënten)
Bij gebruik van allergeenspecifieke immunotherapie moeten de deelnemers een stabiele onderhoudsdosis hebben
Stabiel schema van CF-therapie gedurende ten minste 6 weken voorafgaand aan de screening (alleen CF-patiënten)

Korte samenvatting: Het doel van deze studie is het evalueren van de veiligheid, verdraagbaarheid en farmacokinetiek van ARO-RAGE in normale gezonde vrijwilligers (NHV's) en in deelnemers met inflammatoire longaandoeningen. ziekte (astma, cystische fibrose [CF]). In deel 1 van de studie krijgen NHV's een enkele dosis ARO-RAGE of placebo. In deel 2 van de studie zullen volwassen deelnemers met inflammatoire longaandoeningen 2 doses ARO-RAGE of placebo ontvangen. Extra NHV's kunnen worden gerandomiseerd om 1 of 2 doses ARO-RAGE of placebo te ontvangen naar goeddunken van de sponsor. Dosisniveaus in deel 2 zullen worden bepaald op basis van cumulatieve veiligheids- en farmacodynamische gegevens van deel 1.

Wat ik leuk vind aan ARO-RAGE is dat het niet specifiek is voor een longziekte, maar potentieel breed toepasbaar is met nieuwe MOA.
Relatief hoog biologisch risico, extreem hoog medisch/financieel potentieel. $100 miljard aan toekomstige MC's komt goed uit...

Helaas, toch op de valreep beetje bijzonder nieuws !
Toch is het voor dagen als vandaag dat ik per $ minder al kooporders durf te plaatsen.
Zelf de $26,35 ook net gemist en dat voelt erg dubbel.

Inderdaad weer wat moois met Rave om over te dromen.

rage appears to be the gift that keeps on giving: RAGE is clearly an important inflammatory mediator in many pulmonary diseases and is an attractive therapeutic target.

sRAGE normally circulates in the blood at low levels, and sRAGE levels increase in patients with inflammatory diseases, highlighting a potential role for sRAGE as a biomarker [143]. Administration of sRAGE as a therapeutic agent to block RAGE signaling has shown promising results in studies of asthma [80], chronic hypoxia [144], and cystic fibrosis [90]. Further work here in clinical trials is warranted.

Other methods of blocking RAGE specifically in the lung have not yet been tested. These other inhibitors, such as anti-RAGE antibodies and small molecule inhibitors of RAGE (pyrazole-5-carboxamides; TTP488, azeliragon from Trans-Tech Pharma, LLC; FPS-ZM1) have shown promise in other tissues and disease models [24,145–149] and are just beginning to make their way into human clinical trials for treatment of Alzheimer’s disease

Het opnemen van CF patiënten in de RAGE trial komt niet uit de lucht vallen. Citaat uit voornoemd artikel:

Further research into the timing of RAGE’s role in asthma pathogenesis has shown that it can act as early as the sensitization phase of allergic airway responses. Ullah et al. found that, during HDM- or cockroach-induced AAI, airway sensitization to allergens relied on intact RAGE signaling and was mediated by the RAGE ligand, HMGB1 [81]. Another group demonstrated a role for RAGE in T cell activation in OVA-induced allergic airway sensitization [82], suggesting that RAGE is important in both the early innate and adaptive immune responses to allergens.

More studies are needed to fully characterize all of RAGE’s effects in the complex pathways that contribute to asthma pathogenesis, but at this point in time, RAGE does appear to be a key early mediator of allergic airway responses in both mouse models and human patients.

Cystic fibrosis

Polymorphisms of the AGER gene have been associated with increased disease severity in patients with cystic fibrosis (CF). AGER -429T/C is associated with decreased FEV1 and increased AGER promoter activity, leading to increased RAGE expression and worsening lung function in CF patients [89]. The AGER -374T/A variant was also associated with increased RAGE expression and resulted in increased IgE levels in the lungs of CF patients [90]. The authors hypothesized that CF patients with this RAGE-mediated increase in allergic airway inflammation were more susceptible to sensitization by environmental allergens and pathogens such as

COPD is a heterogeneous respiratory disease associated with exposure to various pollutants, most notably cigarette smoke, resulting in a chronic inflammatory response leading to airflow obstruction. The RAGE axis appears to play a role in the development of COPD. Enhanced expression of RAGE and its ligands [19,128–135], and decreased sRAGE expression in the lungs [135–140] suggests RAGE signaling contributes to the chronic inflammatory response in COPD. Activation of RAGE signaling in response to smoke likely initially evolved as a protective mechanism to promote an inflammatory response against inhaled pollutants; in smokers who are constantly exposed to smoke, however, there is a “tipping point,” at which this response goes unchecked, possibly due to genetic susceptibility factors [141,142]."
